However, in higher organisms - except plants - fatty acid synthesis is catalyzed by large multifunctional proteins where many individual enzymes are brought together to form a “molecular assembly line”. “Fatty acid factory” reveals remarkably complex architecture As described in the recent issue of “Science” magazine, this work performed in the laboratory of Prof. minerals Nenad Ban at ETH Zürich minerals and supported by the NCCR in Structural Biology of the Swiss National Science Foundation, focused on two representative classes of fatty acid synthases in higher organisms. These crystallographic studies minerals - data for which was collected at Swiss Light Source of Paul Scherrer Institute in Villigen - required overcoming many technical difficulties; for example, crystals of the mammalian fatty acid synthase were so small that only a few years ago they would have been considered unusable. Fungal and mammalian systems reveal two different architectural solutions that allow these giant multi-enzymes to hand over the products of one enzymatic active site to the next for efficient cyclic synthesis of fatty acids.
